Usually not by itself. Sanitizing is a step after cleaning, never instead of it. An antimicrobial sprayed over loose dust just bonds to the dust. The product also has to be EPA-registered and labeled for HVAC interiors, applied at the label dwell time. Ask to see that label.
Material decides what is possible. Rigid galvanized trunk lines in 1950s–70s attic systems take a wipe-down and a direct application well. Flex duct liner and fiberglass-lined plenums are porous. Late-summer wildfire soot settles into that fiber and holds the smell. No antimicrobial pulls it back out. Replacing that run is the honest answer.
Mold needs a water source. Condensation staining on a supply plenum after marine-layer nights means a cold uninsulated face or a gap in the wrap. Treat the surface and the stain comes back next season. Rodent or bird nesting in attic ducts follows the same logic: remove nest material and droppings, repair the failed boot or screen, then sanitize.
Panned joist returns in older tract homes are framing cavities, not sheet metal. Raw wood and old dust cannot be sanitized in any meaningful way. Sealing and converting is the real fix.
Plan to be out of the house during application and through the dwell period.
